報(bào)告摘要:

We present a physiologically grounded mean-field model of sleep-wake regulation that unifies wakefulness,  NREM ,andREM sleep within a single dynamical framework.Extending the Action-Arousal  architecture, we explicitly incorporate wakefulness-specific populations (Action and Arousal) alongside NREM and REM populations, and we embed circadian, ultradian, homeostatic, and REM-pressure drives into the model.Dynamical analysis using a potential-landscape theory and linear stability analysis reveals multiple stable attractors corresponding to wake, NREM, and REM states. This framework clarifies how parameter variations reshape transition thresholds. Notably, wake maintenance is enhanced by reciprocal arousal-action excitation, which also constrains direct WaketoREM transitions. In contrast, the NREM-REM cycle appears largely insensitive to reciprocal arousal-action excitation, indicating a modular separation between wake stability and the core NREM-REM oscillator. Our formulation reproduces key features of human sleep architecture, including robust NREM-REM ultradian cycling (~90 minutes) embedded within a broader ~24-hour sleep–wake rhythm, with realistic transition sequences and hysteresis between states. The framework offers a versatile, testable platform for exploring sleep regulation, with potential translational relevance to insomnia, hypersomnia, REM sleep disorders, and chronotherapy.
